He is best known for his appearances on Comedy Central's televised roast specials, as well as his work on the network's television shows Tough Crowd with Colin Quinn, Lewis Black's Root of All Evil, and the Stand-Up Nation programming block, which he hosted.

How Did Greg Giraldo Die?
Giraldo overdosed on prescription medication on September 25, 2010; the precise substance is unknown. It is thought to have been an accident. He had been supposed to perform at the 3rd Annual New York Recovery Rally earlier that afternoon, introducing singer Courtney Love.
The purpose of the event, which took place on Randalls Island in New York City, was to “celebrate the reality of addiction recovery and offer hope to those who have yet to find recovery.” Giraldo was supposed to perform between 12 and 3 p.m., but he never showed up.
When Giraldo failed to show up for a scheduled concert at The Stress Factory that evening, police were called, and officers rushed to his room at the Hyatt Hotel in New Brunswick, New Jersey, where they discovered him motionless and called EMTs.

Giraldo was transferred to the adjacent Robert Wood Johnson University Hospital, where he was kept alive in a coma. At the request of his family, he was taken off life support, and he died on September 29, 2010, at the age of 44.
Giraldo was buried with the inscription “Beloved Father / Always Loved / Forever Remembered” at Good Ground Cemetery in Hampton Bays, New York.

On January 23, 1999, he married his second wife, Maryann. She used to work as a waitress at Caroline's comedy club. They had three sons together.
